The E.coli dipslide measures a total count on one side and coliforms count on the other side. It also reveals E.coli when exposed under UV fluorescent lighting (our UV lamps can be used for this). The E-coli dipslide has two tests, one on either side of the dipslide. One side of the dipslide tests for total count bacteria which gives you an idea of the 'general health' of your sample. The other side of the dipslide tests for coliforms. If colonies flouresce on the coliforms side of the dipslide when exposed to UV fluorescent lighting, these flourescent colonies can be presumptively identified as e.coli (details of our UV light are further down this page). If your test sample contains microbiological contamination, colonies will grow on the dipslide. You would then compare the amount of colonies with the chart provided to assess the level of microbes present in the sample. These dipslides come in a pack size of 10 slides per box. The slide has a flexible paddle and is within a shatterproof container. To get accurate results, you will need a long wave 366nm UV light / lamp. Remove the dipslide from its container. Be careful not to touch the agar while doing this. 2. Dip the dipslide in the sample for approximately 10 seconds. 3. Allow the the fluid to drain from the dipslide. 4. Place the dipslide back within the container. Seal firmly. 5. Label the dipslide using the included labels. 6. Place the dipslide within the incubator for 24-48 hours at 35 degrees C. 7. Once incubated, compare the amount of colonies on the dipslide with the chart included. The straw coloured (plate count) side will indicate how many viable organisms are within your sample. The red (MacConkey 3) side will indicate the level of coliforms in your sample. Red / pink colonies on the MacConkey 3 side indicate coliforms. The chart will help you assess the level of total count / coliform contamination. 8. To check for e-coli colonies, examine the MacConkey 3 side under the uv lamp. Turn the light out / examine in the dark so that it is easier to view. 9. Red / pink colonies that fluoresce blue / green under UV light can be presumptively identified as e.coli colonies. Colonies that do not fluoresce indicate a negative result. Colonies that are not red / pink that fluoresce also indicate a negative result. Surface testing 1. Remove the dipslide from its container. Be careful not to touch the agar while doing this. 2. Holding the tabs at both ends, press one side of the dipslide onto the surface that you are testing. Press down gently ensuring full contact with the surface. 3. Repeat on the other side of the dipslide. 4. Place the dipslide back within the container. Seal firmly. 5. Label the dipslide using the included labels. 6. Place the dipslide within the incubator for 24-48 hours at 35 degrees C. 7. Once incubated, compare the amount of colonies on the dipslide with the chart included.
